Two Secretagogues Acting on Two Different Receptors
This vial contains 5mg of ipamorelin and 10mg of tesamorelin dried together, 15mg total net peptide, and the pairing is built on the fact that the pituitary has two independent switches for growth hormone release. Tesamorelin is a GHRH analog: it binds the growth hormone-releasing hormone receptor on somatotroph cells and drives synthesis and release through the canonical pathway. Ipamorelin is a growth hormone secretagogue receptor agonist — it binds the ghrelin receptor, a different class of receptor on the same cells, and works partly by suppressing somatostatin, the inhibitory hormone that clamps growth hormone release between pulses. Because one pushes the accelerator and the other releases the brake, the two are studied together rather than as alternatives. Ipamorelin is also the most selective of the older secretagogue series: unlike GHRP-6 and GHRP-2 it shows little effect on cortisol or prolactin, which is why it displaced them in most protocols. Both components are peptides and both are subject to the same handling constraints once the vial is reconstituted.
