NNMT Inhibition and the Methyl-Donor Sink
5-Amino-1MQ (5-amino-1-methylquinolinium, supplied as the iodide salt, C10H11IN2, molecular weight 286.11 Da) is a small-molecule inhibitor of nicotinamide N-methyltransferase, an enzyme that sits at an awkward junction between two metabolic pools. NNMT takes nicotinamide — the salvage precursor cells use to regenerate NAD+ — and methylates it using S-adenosylmethionine as the methyl donor, producing 1-methylnicotinamide, which is excreted. Every molecule that goes down that route is a molecule of nicotinamide lost to NAD+ salvage and a molecule of SAM spent without contributing to methylation of DNA, histones or proteins. NNMT is highly expressed in white adipose tissue and liver, and its expression rises in obesity, which effectively enlarges the sink. 5-Amino-1MQ binds the substrate site rather than the SAM site, with a reported IC50 of 1.2 micromolar, and is described as selective in that it does not meaningfully inhibit related methyltransferases or other enzymes of the NAD+ salvage pathway. Published work reports reduced lipogenesis in 3T3-L1 adipocytes at an EC50 near 30 micromolar and efficacy in diet-induced obesity, muscle injury and ovarian cancer metastasis models in mice.
