Direct Pan-AMPK Activation via Phosphatase Inhibition

O-304, also coded ATX-304 (CAS 1261289-04-6, C16H11Cl2N3O2S, molecular weight 380.25 Da), is a benzothiazole small molecule developed at Umea University and Betagenon AB as a direct activator of AMP-activated protein kinase. AMPK is the cell energy sensor: it is switched on when the AMP-to-ATP ratio rises, and once active it shuts down anabolic pathways that consume ATP — fatty acid synthesis, cholesterol synthesis, protein synthesis via mTOR — while switching on catabolic ones that generate it, principally fatty acid oxidation and glucose uptake. Most compounds that raise AMPK activity do so indirectly by interfering with mitochondrial respiration and letting AMP accumulate; metformin works broadly this way. O-304 is described as acting differently, inhibiting protein phosphatase 2C, the phosphatase that dephosphorylates and thereby switches off AMPK at Thr172. Blocking removal of that phosphate keeps AMPK active without requiring an energy crisis to trigger it. It is called a pan-activator because it acts across AMPK isoform combinations rather than selecting one. Published work reports protection against cisplatin-induced acute kidney injury and improvement in metabolic liver disease models.
